Comparison of Antidepressant Efficacy and Toleration of Sertraline and Amitriptyline in Depressed Patients

Abstract
Comparison was performed to evaluate the antidepressant efficacy of sertraline. A selective serotonin reuptake inhibitor and amitriptyline. Forty-eight patients with DSM-lll-R defined major depression or dysthymia were admitted to the study.
After
one
week washout period patients randomly received sertraline (N=25) or amitriptyline (N=23) for 6-week study period. The mean final daily medication dose was 108mg for the sertraline and 151mg amitriptyline treatment groups respectively.
Efficiency was measured on Hamilton Rating Scale for Depression (HAM-D), Clinical Global lmpression Scale (CGl) and self rating of Beck Depression lnventory (BDl). Significant reduction in HAM-D and BDl from base to the end of treatment groups.
The
HAM-D score was reduced by half or more for 76% of patients taking sertraline and 78% taking amitriptyline. CGl score of 1 or 2 at the final visit was 80% of patients taking sertraline and 78% on amitriptyline. The sertraline group had a higher
proportion of gastrointestinal side effects somnolence and headache. The amitriptyline one showed a higher proportion of anticholinergic side effects somnolence tremor and dizziness.
